Done it before and its a really boring drive if nothing else !
like others have said It depends where you start and Def take the Chunnel !
actual drive time from France is a V boring 6 ish hours (depending on your fear of losing your licence)
+ remember that Dutch public transport is very reliable and very fast.
Have fun anyway, I am sure it would be a laugh if nothing else.

I did this trip back in february this year in my old S. Managed to do it door-to-door in just under 6 hours (London-Amsterdam). Left at 9pm and was knocking on my friends door at 3am(UK time) . Was a hassle-free journey but would recommend that you fill up your tank at Dover, petrol is slightly more expensive on the other side! Unfortunately though, you will lose

Ok, so everyone says ill lose, but j20suf reckons 6 hrs door to door, which is good going. Starting point would be in North London. Probably either mine or my friends house. I would drive off as they walk to a bus stop, then get a train to whichever airport.
I think realistically, to stand any sort of chance i would have to be very well organised and they would have to not be, and preferably i'd prefer if they didnt leave from Heathrow 'cause it's got good transport links. actually, i'd hope they leave from Gatwick.
Either way, it would be great fun win or lose.

taking the train would give you a bit of benefit but i still doubt you'll get there within 6 hours.
North London to Dover is quite a drive and they'd be on the plane before you even reach Dover!

Take the chunnel and make sure that you choose the depart time to coincide with your plans.
When TG started doing this challenge they didn't use flights for the closer destinations. So a fairer competition would be for them to take the tube through London to Kings Cross, Chunnel to France or Belgium then regional and local trains. Being Amsterdam you should of course insist that the last leg of their journey is done on one of the Amsterdam free/rent bicycles. Should make it more interesting.
