Rear Diff. Question

If I jack up my car on one side and turn the rear wheel by hand while the other rear wheel is on the ground in stationary, would this damage the rear diff.?

It might if your diff is made out of toothpicks and candycanes. j/k
Really though, it sounds like you are relating the dangers of when a wheel picks up off the ground while racing (with this type of LSD), and the damage it can cause.
This damage actually happens when the wheel hits the ground again, and the lsd has to work against the momentum of the now spinning drivetrain assembly in an instant.

spider, you'd be OK, but if you are worried, put the trans in neutral like XTC mentioned and the diff won't know the difference between spinning the driveshaft vs the other wheel.
