With a MY2000, drain it. It's time.
I've found if you let it run out of the radiator overnight, most of the cooling system will empty itself. After pulling the block drain, a tablespoon at the most came out. So do the rad only. The drain is easy to get to, look straight down between the fans.
So....if you look inside the radiator with a light, and you don't see any white-ish scale, and it looks clean, refill it with Type2 Honda premix. Don't flush it. Most water will do more harm than good by starting the corosion process.
Top off with Type2 as well if you're not going to change it. But change it

HOLD ON! And Billman can slap my pee pee if I'm wrong.
A year 2000 S2000 does not have Type II coolant. It was the only year that didn't. Plus, it was only good for about 3 years, unlike the original Type II in the later cars, that was good for 10 years (so Honda claims).
Do as Billman says about the change. It's time and long overdue. If you are going to change to a "long life" Type II, it might be wise to flush it well as the two are not compatable with each other.
If you are only going to top it up till you get a chance to change it out, then just add water for now or use "regular" (green, non-premixed) coolant (NOT the long life stuff - Honda Type II or the aftermarket orange stuff).

Good point....My only concern is with putting water in the system, as it starts corrosion. If that's the case, some Polland Spring will make a nice flush
now let me quote Xv: HOLD ON AGAIN!
The reason I came back here to post....the s2k cooling system is very difficult to bleed. Even if you follow the helm's to the T, you will still have air in the system, and wind up overheating the car. I have my own methods, but I would recommend vaccuum filling the system.
If your heat blows hot, you know you got the air out

I used distilled water for the flush.
Yes, it's a bugger to bleed but the Helm instructions seem to work well if followed. I didn't have to add much coolant after the system "burped" itself and I got heat immediately. I also disconnected the coolant line at the oil filter. No way I was going to get that block heater unscrewed.

Like I said, MY2000 was the only S2000 that did NOT have Type II coolant. For MY2001 and later, Honda started to use the longlife stuff.
See Helm manual (2001 S2000 service manual) pages 10-8 to 10-11.
