Is 300whp on the motor realistic?
I am thinking of doing an all-motor build up S. Would 300whp be realistic with a stock bottom end. What about 400whp on built internals race gas all supporting mods?

But don't let me deter you, if that's the way you want to go. Find an engine builder that thinks they can do it, and get a written contract. If they fall short of the goal, then you don't pay (or at least don't pay the full price). Also, if reliability is a concern to you, you may want to keep in mind that a motor like this would probably not last very long.

That's not the only reason. I'm sure this motor is not easy to drop into the usual FWD drag racing chassis (like the Civic HB). And, the S2000 - when stripped down - is probably still heavier than a comparably lightened Civic HB.
However, I think the motor is probably superior to the Civic/Integra motors that are often used in drag racing. But, you'll notice that none of those that are used in competition still have the stock bottom ends. Most of them have been bored or stroked to ~2.0L. You could probably get a stroker kit for the S2000 and bump your power-making potential. Or maybe the new 2.2L engine would have the potential to make 300whp with the right mods. No one really has any idea how responsive to mods it will be, compared to the 2.0L engine.

I think close to 300 with a bore to 2.1 liters
Header back 70mm exhaust (Amuse R1000): 15
Headers: 7
High profile cams with higher redline (vtec killer?): 15
ITB/AEM tuning (as per LHP): 20
13:0 compression (Based on Sideways' est. 6 hp/ 0.5 points): 24
Port and polish (Based on Sideways' results): 10
2.1 L bore (proportional increase to displacement): 10
=+101 HP.
I could be WAY off though, and some of these gains may be max gains, not peak. So perhaps, conservatively 280?

Saiyajin- You are taking the max gains that people have listed for each of those mods and adding them up. That's not necessarily the way that mods go.
For example, if you add a Toda header to a stock S2000, you may get 7 hp. But if you add it to an S2000 that already has an Amuse exhaust, it may only net an additional 4 hp (over the power that the exhaust added). However, it may also be possible that it gives you 10 hp instead of 7. It's sometimes tough to tell. Some mods may make other mods more/less effective.
Also, your mods require going into the bottom end, and slow4dr said he didn't want to do that.
