It was when the car was built in the Tochigi plant. Since they moved production out of there to a differnt plant, I think it's made by plain old robots now just like the Accords.

Dunno about the Suzuka plant, but yes, all S2000s made in the Tochigi plant are put together by hand. (Although the parts themselves are not hand-manufactured, of course.)

What difference would it make? Would you enjoy the car any more/less if it was hand assembled (or not)?

It can be good or bad. As a prior owner of a TR6, the last hand-built British Roadster, it was remarkable how different each car was. The tolerances for gaps and seams were non-existant. Different fastners were used on comparable parts. I guess that is the bad. On the good side, there can be a level of quality that is not present in a mass-manufactured car. This assumes the assemblers are checking and rechecking what is taken for granted when a robot assembles the same thing over and over.
